Pharmacological Profile of Garcinielliptone FC from Platonia insignis

Abstract

Platonia insignis Mart., Clusiaceae, or the bacuri tree is found in the Amazon rainforest and several countries of South America. The tree is known for its solid timber, its edible fruit, and some derived products such as the bacuri seed butter used in phytotherapeutic preparations and cosmetics. Many bioactive natural products have been isolated from the bacuri tree, in particular the polycyclic prenylated acylphloroglucinol–designated garcinielliptone FC, initially isolated from Garcinia subelliptica Merr. (like the 32 garcinielliptones known to date), but it can be isolated also from the seeds and fruits of bacuri. We have reviewed the pharmacological properties of this under-exploited natural product which displays antioxidant, vasorelaxant, anticonvulsive, antiparasitic, and anticancer properties, all discussed here. The marked antiproliferative activities of garcinielliptone FC against various cancer cell lines, and the structural analogy with tumor active prenylated acylphloroglucinols, such as hyperforin and nemorosone, argue in favor of further in vivo evaluations of this active metabolite against solid tumors and leukemia. By analogy with other prenylated acylphloroglucinols, the chaperone protein HSPA8 is proposed as a molecular target for garcinielliptone FC. The compound is recognized as being safe and well-tolerated in rodent species according to toxicology studies. Altogether, the review shed light on the pharmacological interest of garcinielliptone FC and bacuri fruits.
