Malate is a dicarboxylic acid that plays an important role in the Krebs Cycle. It is an intermediate that forms from fumarate in the Krebs Cycle.

Malate dehydrogenase (MDH) is an enzyme which reversibly catalyzes the oxidation of L-malate to oxaloacetate in the presence of NAD.

Fumarase is an enzyme that catalyzes the reversible hydration/dehydration reaction of fumarate to malate.

Isocitrate is a substrate in the citric acid (TCA) cycle and is formed by the isomerization of citrate. It is oxidized by isocitrate dehydrogenase (IDH) producing alpha-ketoglutarate and generating NADPH.

L-malate (L-malic acid) assay kit

Assay Genie’s L-malate assay kit is based on malate dehydrogenase catalyzed oxidation of malate in which the formed NADH reduces a formazan (MTT) reagent. The intensity of the product color, measured at 565 nm is proportional to the malate concentration in the sample.

The increase in absorbance at 565 nm is proportional to the enzyme activity.

Fumarate assay kit

Fumarate (Fumaric Acid) is one of the key components in TCA cycle and is used by cells to form ATP.

This fumarate assay kit is based on fumarase catalyzed hydration of fumarate to malate. The malate is then oxidized by malate dehydrogenase generating NADH which reduces a formazan (MTT) dye. The intensity of the product color, measured at 565 nm is proportional to the fumarate concentration in the sample.

The converted dyes have an absorption maximum at 565 nm. The increase in absorbance at 565 nm is directly proportional to the isocitrate concentration or IDH activity.
